"N Keyrollover" has been a buzzword when promoting modern mechanical keyboards. So...
Has [lack] of Key rollover ever affected you?
I've been hard pressed to find a situation where 2KRO wasn't enough.
If it has been a problem feel free to list Keyboard/Game/and Combination it failed on.

Only time I've been seriously affected is with old keyboards.
I cannot use ctrl + shift + arrow keys (Excel) on my Suntouch Jr.
In some fast FPS, 2KRO can cause a problem with advance button combinations. (GunZ comes to mind, or if you BHop or use any animation cancels in games)
Fighting games and multi player 1-keyboard games are another example.

I was playing CS:GO with my Siig Minitouch. I couldn't strafe and run forward at the same time. That was the first time I noticed rollover issues. Other than gaming, I've never really noticed it.

Yes, FPS gaming on my Microsoft Sidewinder X6... discovered it's got 2KRO after some weird behaviour, crouching and moving I coudn't reload... kind of critical in the specific situation. And not good on a board marketed to gamers...

If I swap Ctrl and Caps Lock on my G84-4100, caps lock will randomly turn on if I press keys on the right side of the board in quick succession. Easy to get around by just making both keys Ctrl, but it's still annoying and I suspect 2KRO is the culprit.
Otherwise, no. I used a combination of Model M (regular use) and Data911 (gaming) for over six months without problems, and both are only 2KRO.

I find it funny that a lot of people use their "N-key rollover OMFGBBQ" boards thru usb and not ps2 to fully embrace the N-key rollover powers of their mechs and settle with 6KRO instead.

I used to run into a lot of issues, when playing retro multiplayer games on one keyboard with friends. Or even when playing Bomberman or some racing games alone on arrows (yes, I admit I've always preferred arrows to WASD/ESDF/whatever).

...unless the keyboard supports NKRO over USB, right?

I find 2KRO to be insufficient for gaming (tried gaming on my Model M for a bit and I found that I had an inability to run at an angle and reload), but I've never had a need for more than the standard 6KRO over USB.

I had some issues with the minitouch and the cherry G84-4100 I have.
The 4100 has some of the worst blocking / ghosting issues I've yet seen.
Though given how compact the matrix is, I think the WYSE may be worse. I haven't tried it yet.

If you use voice chat and have push to talk on your keyboard rather than mouse you'll encounter issues quite commonly.
If you play any sort of game that uses two hands on keyboard, rather than one keyboard one mouse.
Otherwise, zero issues.

6kro is sufficient more often than not, but the thought of having the option of nkro is alluring if it were to ever be truly necessary. better safe than sorry!

Just thought I would add, I am very happy with 6KRO since the time you need to mash a bunch of buttons at the same time (gaming) I usually only have one hand on the keyboard and the other on my mouse, so I can only press 5 buttons at once.

Mainly FPS and never got hit by 2K rollover on my M.
I'm left handed and only use my right hand over
arrow keys
right(CTRL, SHIFT, ENTER)
Ins, home, P-Up, del, end P-Down
and the num pad
I'm sure I frequently pressed more then 2 keys at the same time but it never got affected by combos from the keys listed above.

I can't play dota with 2KRO. Get a bunch of problems. 6KRO is fine for everything though.

Well playing Planetside 2, I had a keyboard that had 4KRO.... When I would fly the Mosquito (http://img2.wikia.nocookie.net/__cb20120930204027/planetside2/images/d/dc/Vehicle_Mosquito.gif)
And to do a certain maneuver I need to use S, shift, space, and either A or D depending on the direction I want to kinda make it move to. That should be enough keys right? Well, yes. But the matrix if I press S and shift, it won't do it properly lol.
6KRo is enough though.

Nope.
I do game a bit on my Model M, FPS mostly, but it doesn't seem to have affected me.
Most gaming is on Ducky or HHKB though.

I managed to get an old Chicony board working once and I really struggled to play any sort of games with 2KRO. Also, Futaba switches feel weird.

I have a lot of different key setups for different game genres, and on top of that, my typing style is a bit "slurred" (meaning I tend to err on the side of holding keys longer, so I'm often still holding previous key(s) while pressing the next). As a result, 2KRO tends to be a problem for me, though some 2KRO designs are much worse than others.
My MicroConnectors D07-135, for instance, had big issues in FPS, because it didn't like W + E + Space (forward-sprint-jump). I-Rocks KR-6260, a dome keyboard with a "gaming optimized matrix", would reliably block out the "r" in "keyboard" during fast typing. I don't use my Model M enough to recall exactly what blocks and what doesn't, but I remember hitting the limit once or twice.
Now with 6KRO, I find that it's vastly less common to run into issues. It's almost always fine for recent games, where the mouse is used as a main form of input, but for older games and keyboard piano, occasional trouble can occur. And of course, it's always nice to have immunity to my old trick for winning at games with multiple players at the same computer: wait for a critical moment, then slap the nearest available section of the keyboard with your whole hand to block out the other guy's input.
